Four Levers on One System

NAD⁺, methylene blue, SS-31, MOTS-c — people lump them all together as "mitochondrial support" and assume they're interchangeable, or all "cleanup." They're not. Each acts on a completely different level of the mitochondrion, and only one of them actually cleans. Sort them properly and you'll know exactly what each is buying you — and why they stack instead of overlap.

The confusion is understandable: four agents, all described as "good for mitochondria," all in the same conversations. But "good for mitochondria" is like saying four things are "good for a car" when one is fuel, one is a tune-up, one is the chassis, and one is the fleet manager who buys new cars and scraps old ones. They don't compete — they occupy different jobs. Here's the model that separates them cleanly: fuel, flow, structure, fleet — four levels of one system, one agent at each.

What each one actually does

Level 1 · Fuel

The currency the chain runs on — the electron carrier that shuttles the energy of your food into the mitochondria. It doesn't optimize or clean anything; it's the raw material everything else depends on. Also powers sirtuins and DNA-repair enzymes, so it enables repair and signaling without doing them itself. Not cleanup — it's the fuel.

Level 2 · Flow

Manages electron traffic through the existing chain — carries electrons past a sluggish Complex I, upregulates Complex IV, and recycles NADH back to NAD⁺ (so it directly interacts with the fuel level). It makes each engine run better right now. Not cleanup — it's a performance tune, a protector of function.

Level 3 · Structure

Binds and stabilizes cardiolipin — the fatty scaffold of the inner membrane that the whole electron transport chain is embedded in. Keep the scaffold organized and the complexes sit correctly and leak less. It fixes the environment the machinery lives in. Not cleanup — it's structural protection.

Works above the single mitochondrion, via AMPK: signals biogenesis (build new mitochondria) and mitophagy (clear damaged ones), plus metabolic flexibility. This is the only true cleaner of the four — it renews the population rather than tuning any single unit.

The real question So which of them "clean"?

It's worth asking directly whether these are cleanup agents or something else. Now the answer is crisp — and it corrects the most common misconception in this space:

The correction most people need

Three of your four "mitochondrial" agents are not cleanup at all — they supply, optimize, or protect. Only MOTS-c actually clears damaged mitochondria. So if you were counting NAD⁺, MB, and SS-31 toward your cleanup gap, they don't fill it — your cleanup still rests on MOTS-c plus the behavioral levers (fasting) and the oral cleaners (Urolithin A, spermidine). Don't let "mitochondrial support" masquerade as "mitochondrial cleanup."

The important limit

What none of them do: fix a broken complex

Here's the subtlety worth holding. None of these four repairs a damaged Complex I or IV protein directly. NAD⁺ feeds it, MB routes around it, SS-31 protects its surroundings, MOTS-c replaces the whole unit. The only ways the machinery itself actually gets fixed are: replacement (biogenesis builds brand-new complexes — driven by your training and MOTS-c), protection (SS-31, glutathione, CoQ10 keep them from degrading), and cofactor support (B2 for Complex I). "Repair" of a mitochondrion is really renewal plus protection — not component-level patching.

Why they stack — and where they sit in the framework

Because each occupies a different level, combining them optimizes fuel, flow, structure, and fleet simultaneously — that's the real reason the "supercharge" happens. In your verb framework: NAD⁺ = supply, MB = protect/optimize, SS-31 = protect, MOTS-c = clean+build. Notice the balance — three keep the current system running well, and only MOTS-c renews it. If you want the system to get younger rather than just run better, the fleet level (MOTS-c + training + your cleanup work) is the one to prioritize.
